傳統(tǒng)工藝:
我國首創(chuàng)了二次發(fā)酵法生產維生素C,全工藝包括發(fā)酵、提取和轉化三大步驟,即以D-葡萄糖為原料,通過兩次發(fā)酵生成2-酮基-L-古龍酸,經提取后用酸或堿轉化成維生素C,其先進性得到了國際公認,但在發(fā)酵液中殘留著菌絲體、蛋白質和懸浮微粒等雜質。原工藝采用加熱沉淀法去除這些雜質,即將發(fā)酵液靜置沉降后通過離子交換樹脂,部分酸化2-酮基-L-古龍酸鈉,調節(jié)PH值至蛋白質等電點并加熱使蛋白質凝聚,然后用高速離心法分離出凝聚的菌絲、蛋白和懸浮微粒,清液再次通過離子交換樹脂,全部酸化為2-酮基-L-古龍酸水溶液,在進行濃縮結晶,其工藝流程為:
發(fā)酵液—沉降—樹脂調pH—加熱—冷卻—離心機—樹脂脫鹽—濃縮結晶—2-酮基-L-古龍酸
此工藝通過一次樹脂和加熱沉淀法除蛋白,加熱既要耗能又造成2-酮基-L-古龍酸損失近4%,且發(fā)酵液直接通過離子交換樹脂,是樹脂表面污染嚴重,交換容量下降。
結合膜分離工藝:
發(fā)酵液—超濾—樹脂脫鹽—濃縮結晶—2-酮基-L-古龍酸
超濾截留分子量范圍從500到50萬,而維生素發(fā)酵液的分子量范圍一般在1萬到10萬,所以選擇一定截留分子量的超濾膜可以除去蛋白等雜質。
VC膜分離工藝的優(yōu)點:

2、節(jié)約能耗,提高收率,一般收率可達99%
此工藝采用超濾法一步去除發(fā)酵液中殘留的菌絲體、蛋白質和懸浮微粒等雜質,省略了預處理、加熱、離心等工序,既節(jié)約了能耗,又提高了古龍酸的收率。由于超濾是一個物理過程,它本身不會造成2-酮基-L-古龍酸的分解或變化,收率主要取決于濾渣中含2-酮基-L-古龍酸的量,而超濾的濾渣中含2-酮基-L-古龍酸的量小,且可通過加水洗滌,所以超濾收率可達99%。
